UW Microbiology has an excellent opportunity for a Research Scientist 1.
The research that this position is engaged in are studies which develop and test novel vaccines and immunomodulators for the therapy or prevention of infectious diseases with an emphasis in HIV and influenza research. Projects include, but are not limited to, investigating the role of virus specific T cell responses and antibody responses in viral control; testing immunogenicity and efficacy of vaccines and therapeutics in nonhuman primate models for HIV, ZIKV and influenza; investigating mechanisms underlying mucosal dysfunction induced by lentivirus infections; development and treatment of the latent viral reservoir during antiretroviral drug therapy; and novel therapeutic interventions to treat HIV disease pathogenesis.
RESPONSIBILITIES
• Tissue culture techniques.
• Preparation of necropsied tissues.
• Tissue processing, including isolation of lymphocytes from lymph nodes, jejunum, vagina, rectum, blood, colon, and bronchio-alvelolar lavage using Percoll, Ficoll, and other lymphocyte separation techniques.
• Cryopreservation of cells.
• Support for immunophenotyping and intracellular cytokine staining assays.
• ELISA (enzyme-linked immunosorbent assay).
• Managing, organizing, and inventorying a tissue repository.
• Shipping category B samples to collaborators.
• Management of research reagent and supplies including stocking of BSL3 retrovirus labs.
• Maintain electronic laboratory notebook.
• Regularly report experimental results to manager.
• Present results in lab meetings.
• Perform biosafety 2-3 practices.
• Perform flow cytometric analysis of cell mediated immune responses.
• Functions as a backup to the postdoctoral staff.
• Receive/store shipments from collaborators/vendors.
• Monitor supply inventory and alert purchasing team when low.
• Coordinate broadly with team members to prepare equipment/supplies in support of tissue processing/assays.
MINIMUM REQUIREMENTS
• Bachelor's degree in microbiology or related field.
• Three months to one year of relevant laboratory research experience.
Additional Requirements:
• Experience and knowledge in BSL2-3 practices.
• Experience with specimen preparation from nonhuman primates.
• Experience with cryopreservation of nonhuman primates specimens.
• Working knowledge and experience cell counting using a Nexcelcom cell counter.
• Significant experience in processing tissues from nonhuman primates including purifying lymphocytes from lung, spleen, liver, bronchioalveolar lavages, lymph nodes, GI tract, and reproductive tract tissues.
• Experience in long-term tissue fixation in formalin and ethanol.
• Prior experience with organizing and restocking laboratories.
• Routinely checking research reagent and supply stocks to determine inventory level, anticipating needed supplies; routine inspection and maintenance of laboratory equipment.
• Assisting with organization and maintenance of lab space including organizing newly purchased laboratory supplies; setting up and cleaning up after experiments; autoclaving, aliquoting of reagents; maintenance of a BD LSR II flow cytometer; CS&T of LSR II; acquiring samples on a BD LSR II flow cytommeter and Cytek Aurora cytometer.
• Demonstrated ability to work as part of a team and adapt to different tasks.
• Excellent working knownledge with data analysis, inventory and presentation software including MS Excel, Powerpoint, FACDiva
• Sample retrieval and shipment to collabroators.
Equivalent education and/or experience may substitute for minimum requirements.
DESIRED REQUIREMENTS
• Experience with analysis of CD4+ and CD8+ T cell responses in nohuman primate specimens; Experience with data analysis software including Flow-Jo, Prism, Boolean gating and polfunctionality scoring.
• ELISpot assay performance and analysis; performing various surface, and intercellular cytokine stains for FACS.
• Ability to make independent judgments that will promote efficiency and add value to laboratory's research.
• Strong organizational skills including ability to work efficiently with competing priorities and deadlines.
• Experience with packing and shipment of precious Category A research samples.
